Ryanodine receptors (RyRs) are intracellular calcium channels that are expressed in the endoplasmic reticulum or sarcoplasmic reticulum and which open in response to ligand (Ca2+) binding. Three genes exist encoding RYR1 (skeletal muscle distribution), RYR2 (cardiac muscle distribution), and RYR3 (ubiquitous expression). They are key components of excitation contraction coupling in muscle and defects in their function is extremely harmful. RyR function is controlled by a wide variety of allosteric regulators (calmodulin, ATP, cyclic ADPribose, etc.) and by post-translational modification (posphoylation, nitrosylation). Abnormalities in phosphorylation of RyR2 contributes significantly to cardivascular disease (CPVT, heart failure, arrhythmias).
